「SERAマラリアワクチン実用化のための流行地における疫学研究」

研究概要	本研究課題は、我々の開発したレコンビナントSE47'蛋白質を用いてヒトにおけるSE47'ワクチンPhasel、Phasellトライアルを行うために、その前段階としてマラリア流行地域に居住するヒトのSE47'蛋白質に対する抗体価とマラリア症状との相関を明らかにし効果の予測を行うとともに、ワクチンPhasel、Phasellトライアルの対象集団を特定することである。ウガンダのAtopiParish村では雨季において、10才以下の児童のマラリア感染率が90%以上に達する高度流行地域である。ウガンダ・分子生物技術研究所のEgwang博士との共同研究により、10才以下の児童について、SE47'蛋白質に対する抗体価を測定した結果、抗SE47'蛋白lgG3抗体を持つ児童はマラリアを発病しないことが明らかになった。本年度はさらに解析を進めるため、抗体価測定のコントロールとしてSE47'蛋白質と同様に調整したSE50A蛋白質を用いた。試験管内の実験から抗SE50Aマウス抗体はマラリア原虫の増殖を阻害しないため、格好のネガティブコントロールとなる。10才以下の児童についてSE50Aに対する抗体価は全くマラリア症状と相関しなかった。さらに、検体数を増やし、これら、抗SE47'蛋白抗体価及び抗SE50A抗体価と児童の血中マラリア原虫率の相関を調べたところ、マラリア症状との相関と全く同様に、ほぼ例外なく高い抗SE'47蛋白抗体価をもつ児童は極めて低い血中マラリア原虫率を示した。一方、抗SE50A抗体価として世界各国で研究が進められているAMA-1、EBA175についてもマラリア症状との相関は得られなかったとことから、SE47'蛋白質が最も有望なワクチン候補であることが示された。
