プリオン蛋白欠損マウスにおける小脳ブルキンエ細胞死と脱随機序の解明

研究概要	我々は独自に作製したPrP欠損マウス(Ngsk Prnp^<0/0>)に加齢に伴うプルキンエ細胞死に基づく小脳失調が惹起されることを報告した。本年度は、この神経細胞死からの回復(rescue)を種々のPrP遺伝子のNgsk Prnp^<0/0>への再導入により試みた。用いたPrP遺伝子はアストロサイト特異的GFAPあるいはニュウロン特異的NSEプロモーターにドライブされた正常ハムスターPrP遺伝子(GFAP-HaPrnp及びNSE-HaPrnp)、及びPrP遺伝子プロモーターにドライブされたOcta-repeat欠損マウスPrP遺伝子(MoPrnpΔ23-88)とE200K(家族性CJD型変異)マウスPrP遺伝子(MoPrnpE200K)である。その結果、80週齢までの観察でNSE-HaPrnpは完全にプルキンエ細胞死を阻止した。GFAP-HaPrnpは60週齢時点ではプルキンエ細胞死を阻止していたが、その後振戦が出現したため病理変化を現在検索中である。ちなみにコントロールのNgsk Prnp^<0/0>では45-50週齢でプルキンエ細胞死が明らかとなった。一方、MoPrnpE200Kはプルキンエ細胞死を阻止したが、MoPrnpΔ23-88は阻止できなかった。E200K(家族性CJD型変異)PrPは神経変性阻止に関わる生理機能を保持すること、PrP^Cの生理機能発現にはCu^<2+>結合部位とされるOcta-repeat領域が重要であることが強く示唆された。
