アジアにおける新興・再興インフルエンザウイルスの宿主域変異機構の解明とその制御

研究概要

南中国では、トリインフルエンザウイルスが直接ヒトへ侵入する現象が続いている。本年度の研究により、カモインフルエンザウイルスの宿主域変異機構に関して極めて重要な結果が得られた。
1)カモインフルエンザウイルスの標的器官は腸管であり、特に、腸管のクリプト細胞でウイルスの増殖が見られることを見いだした。カモの腸管の粘膜上皮細胞に存在するインフルエンザウイルス受容体のシアル酸結合様式は2-3タイプである。従って、ヒトから分離されるインフルエンザウイルスはカモに感染できない。カモから分離されるインフルエンザウイルスは、Neu5Ac2-3Gal,Neu5Gc2-3Galの両者に結合できる。筆者らは、カモ腸管のクリプト細胞には特異的にNeu5Gc2-3Gal結合を持つシアロ糖鎖(カモインフルエンザウイルスが結合する受容体シアロ糖鎖)が存在することを見いだした。同時にNeu5Gc2-3Gal β 1-糖鎖への結合性の獲得には、ヘマグルチニン分子内の2つのアミノ酸置換、Leu226→GlnおよびSer228→Glyが必須であることも明らかにした。そして、ヒトから分離されたウイルスへマグルチニン内の2つのアミノ酸置換、Leu226→GlnおよびSer228→Glyを人工的に導入したインフルエンザウイルスはカモに感染・増殖できることを明らかにした。
2)H3亜型ヒトA型インフルエンザウイルスのへマグルチニン分子内のSer228の他に、受容体結合ポケット近傍のアミノ酸(143,155,158番)もシアル酸分子種(Neu5Ac,Neu5Gc)の識別に重要であることを明らかにした。
3)本研究では、アジアで分離されたヒト、トリ、ブタインフルエンザウイルスのシアリダーゼ遺伝子の変異とその標的シアロ糖鎖認識との関連についても検討し、トリ由来のウイルスはN-グリコリルノイラミン酸に対する認識が大きいことなどを見いだした。
