研究概要	本研究は発がんプロモーター作用を構成する二つの要となる事象、「ギャップ結合細胞間連絡(Gap junction intercellular communication:GJIC)の阻害」と「アポトーシスの抑制」の意義並びにそれらの相互関連性について調べることを目的としている。昨年度までに、ras導入肝細胞株や、myc、Bcl-2高発現細胞株(ミシガン州立大学)を用い、カテキン類など、日常物質の癌抑制効果(ソウル大学)や、in vitroでの相互のリンクについて明らかにしてきた。さらに、引き続き個体レベルでの相互関係を検討する目的で、コネクシン(Cx-)32遺伝子欠失マウス(ドイツ、K.Willecke博士由来)やCx43欠失マウス(米国、C.Lo博士由来)の供与を受けた。Cx32遺伝子欠失マウスでは、造血幹細胞数が野生型に比べて少なく、このものを受け手とした脾コロニーアッセイでは、同じ野生型の骨髄細胞から形成される脾コロニーの数が少ないことなど、Cx32の骨髄における機能発現の可能性を初めて見いだした。本年度は、Cx32の造血幹細胞での機能を解析する目的で、Cx32遺伝子欠失マウス骨髄細胞で骨髄再建し、メチルニトロソウレア(MNU)による発がん実験を行った。Cx32遺伝子欠失マウス由来骨髄細胞で再建したマウスにも、野生型骨髄再建マウスでも、MNU処置群には共に胸腺リンパ腫などの血液腫瘍が発症し、その発症時期に差異はなく、頻度にも大きな違いを認めない。ところが、Cx32遺伝子欠失マウス骨髄細胞と野生型の骨髄細胞を同数混合して再建した群に発生した腫瘍を観察すると、その比率は圧倒的にCx32遺伝子欠失マウス由来(7/8)であった。この結果は、Cx32の機能不全が何らかの発がん機構に関わることを示唆する。
