研究概要

ヒト疾病に関与する多くの疾患原因遺伝子についてはその機能解析が進み、疾患の表現型と原因遺伝子の生理機能との関連が明らかにされている。しかし、ゲノム解析や既知の遺伝子との相同性により単離、同定された新規遺伝子の生理機能は未だ不明なものが多い。そこで本研究においては、遺伝子情報から新規機能情報を予測することを試みた。
1.膵臓癌で高率に変異が検出される癌抑制遺伝子DPC4遺伝子のコードするSmad4転写因子がTGF-βファミリーのシグナル伝達分子として機能することが最近報告された。本研究代表者、ヴァンデビルド大学の共同研究者らは、Smadがアポトーシス誘導に、また、細胞外マトリックス産生に関与することを明らかにし、Smad遺伝子の多様な生理機能を示した。現在、Smadファミリーに属する8つの遺伝子の構造をコンピューターにより解析し、新たな生理機能を検索している。
2.軟骨分解活性を有するマトリックスメタロプロテアーゼ(MMP)をコードするファミリー遺伝子のうち、MMP-3、MMP-7遺伝子の機能を軟骨肉腫において検索した。MMP-3はマクロファージに対するケモカイン産生を促し、また、MMP-7はマクロファージからのTNF-αの産生を促し、共にマクロファージの浸潤を促進することを本研究分担者とヴァンデビルド大学の共同研究者が明らかにした。
3.成人T細胞白血病ウイルス(HTLV-I)はヒトT細胞に感染し、細胞を腫瘍化する。本研究分担者らは、このときHTLV-Iの有するTax遺伝子のコードする転写促進因子により細胞周期転写因子E2Fが活性化されることを明らかにした。
4.平成12年3月、ヴァンデビルド大学の共同研究者らを東京へ招聘し、これまでの共同研究成果の発表、研究推進方向の検討を行った。
