抗がん剤タキソールおよびその類縁体の効率的不斉全合成

研究概要	筆者らの研究室では従来の合成法とは全く異なる新しい合成戦略によりタキソールの不斉全合成を1997年に達成した。本研究ではここで明らかにしたタキソール全合成法における反応工程数(約60工程)の短縮および全収率の向上を目的として検討を行った。すでに報告したタキソールの不斉全合成法においては鎖状ポリオキシエステルからα-ハロエチルケトンへの変換に7工程の反応が必要であったが、ここで新たに1,1-ジクロロエチルリチウムを用いるエステル類のアルキル化反応を開発しこの変換を僅か一工程に短縮することを試みた。すなわち、1,1-ジクロロエタンとnBuLiから調製される1,1-ジクロロエチルリチウムをタキソール合成中間体であるポリオキシ鎖状エステルに作用させたところ、第三級アルコールを副生することなく対応するα-モノクロロエチルケトンが高収率で得られることが分かった。さらにこの反応のスケールアップを図ったところ、20g以上の原料を用いた場合でも目的の反応が円滑に進行し、再現よく90%以上の収率で対応するケトンを得ることが可能になった。次いで、ヨウ化サマリウム(II)の存在下、合成された鎖状ポリオキシ-α-クロロケトアルデヒドの8員環環化反応を試みたところ、従来のα-ブロモケトアルデヒドを用いる環化反応と同様に目的の反応が円滑に進行し、対応する8員環状化合物が高収率で得られることを明らかにした。環化反応に関しては、低原子価チタンを活性化剤として用いた場合も良好な収率で8員環状化合物が得られることを最近明らかとし、この方法も大量に基質を扱うことのできる優れた手法である。これにより、上述の反応と併せて反応工程数の短縮および全収率の向上を達成したことに加え、タキソールおよびその類縁体の合成中間体を大量に与える簡便で有用な新しい手法を確立することができた。
