超好熱菌の細胞分裂タンパク質に関する研究

研究概要

超好熱菌Pyrococcus kodakaraensis KOD1株からチューブリンをコードする遺伝子をクローン化を試みた。保存配列に基づき、合成オリゴヌクレオチドプローブを作成し、KOD1株の染色体ファージライブラリーから、遺伝子ホモログを有するクローンを3種類取得した。塩基配列を決定したところ、いずれも、4つの保存モチーフ及びGTP結合領域を有するタンパク質をコードすることが明らかとなった。これらをtubA、tubB、tubCと命名した。tubB、tubCがコードするタンパク質は一次配列から細菌のFtsZに近いチューブリンと考えられた。tubA、tubBそれぞれを大腸菌PAT84株(ftsZ84)に導入し、42℃での生育相補を試みたが、大腸菌での相補はなされなかった。一方、野生型大腸菌にtubBを導入したときに菌の分裂以上が認められた。tubA、tubBを大腸菌で発現し、それぞれを組換えタンパク質として精製した。得られたタンパク質のGTP結合活性、GTPase活性の有無、またそれに伴う金属イオンの要求性を確認したところ、TabAはMg^<2+>依存性のGTP結合活性とGTPase活性を示したが、これらの性質はTabAでは認められなかった。また、細胞分裂の周期調節遺伝子ホモログcdcAを取得し、生化学的性質も検討したところ、真核生物のCDCと同様にATPase活性を有することが明らかになった。分裂の際、DNA複製の中心的な酵素であるDNAポリメラーゼについては構造解析のため、タンパク質分子の結晶化を試み、安定な結晶を得た。現在、構造解析中である。KOD1株のヒストンを組換えタンパク質として取得し、DNAへの安定化効果についても検討したところ、特に高温での構造維持に重要であることがわかった。
