植物細胞における病原菌認識装置の分離とそれらの再構成に関する基礎的研究

研究概要	エンドウ-褐紋病菌(Mycosphaerella pinodes)の相互作用をモデルとして、宿主細胞による病原菌認識と応答機構についての解析から、感染初期における病原菌の認識は先ず宿主の細胞壁で行われ、つづいて下流の原形質膜シグナル伝達系が制御されという仮説に到達した。本研究では、宿主細胞の細胞壁ならびに原形質膜における病原菌シグナル応答性分子についての解析を進めるとともに、エンドウcDNAライブラリーからそれらをコードする遺伝子のクローニングを行った。 1)細胞壁における活性酸素生成酵素cDNAのクローニング細胞壁における活性酸素の生成にかかわるペルオキシダーゼ、ポリアミン酸化酵素の活性がエリシターで活性化し、逆にサプレッサーの処理によって著しく阻害されることが明らかとなった。そこで、病原菌シグナルに対する応答性をさらに詳細に解析する目的で、それらをコードする遺伝子(cDNA)のクローニングを行った。現在、それら遺伝子の感染時、エリシター処理組織におけるに発現解析を進めている。 2)原形質膜シグナル伝達系を構成するシグナル伝達分子cDNAのクローニングこれまでに、エンドウにおける防御応答の発現に原形質膜ポリホスホイノシチド代謝系が深く関与していることが明らかとなっている。本年度は、ポリホスホイノシチド代謝系の鍵分子であるホスファチジルイノシトール1リン酸キナーゼ、ならびにそれらの活性調節への関与が予想されるアクチン、GTP結合タンパク質、ならびにヌクレオシド2リン酸キナーゼをコードするcDNAを単離した。現在、大腸菌におけるタンパク質発現システムを利用し、組み換えタンパク質を作成し、これらを抗原として抗体を作製している。引き続き、(1)シグナル伝達におけるタンパク質-タンパク質間の相互作用についての解析、(2)組み換えタンパク質の再構成実験を進め、植物細胞における病原菌認識とシグナル伝達システムにかかわる分子の機能についてin vitro、in vivoの両系から実証していきたい。
