メタアンフェタミン反復投与によるドーパミンニューロンの機能変化に関する研究

研究概要

平成11年度に腹側被蓋野ドーパミンニューロンではドーパミンに対し感受性亢進が起こっていることを報告した。本年度(平成12年度)は、感受性亢進はドーパミンサブタイプであるD1、D2およびD3レセプターのいずれにおいて起こっているのかを、スライス標本を用い、パッチクランプ法により検討を行った。
1.Wistarラット生後7日齢よりMAP(5mg/kg/日、s.c.)を連続5日間投与しする。コントロール群として同用量の生理的食塩水(生食)を同様に投与した。
2.最終投与より5日後にエーテル麻酔の後に断頭し、氷冷下に腹側被蓋野より急性単離した細胞を用いる。
3.パッチクランプ法により、目的細胞にホールセル記録を行い、電気生理学的手法で、腹側被蓋野ドーパミン細胞を同定し、ドーパミンD1およびD2レセプターアゴニストを投与した。
4.D2 agonistのtalipexoleは生食群よりもMAP投与中止5日後群が大きな過分極を引き起こした。
5.D1 agonistのSKF38393およびD3 agonistのPD128907は生食群もMAP投与中止5日後群においても膜電位に影響を及ぼさなかった。
6.D1 agonistのSKF38393とD2 agonistのtalipexoleの同時投与は、talipexole単独よりもさらに大きな過分極を引き起こし、生食群よりもMAP投与中止5日後群がさらに大きな過分極を引き起こした。
以上より、MAP投与中止5日後において、腹側被蓋野ドーパミンニューロンではドーパミンD1及びD2レセプター両者に対し感受性亢進が起こっている事と、D3レセプターが存在していないか存在しても機能していない事を明らかにした。
