二重のプロモーターに支配されるセリンアミノ転移酵素遺伝子の転写開始機構

研究概要	セリン:ピルビン酸アミノ転移酵素(SPT)遺伝子のプロモーター解析の結果、以下が明らかとなった。二つの転写開始部位のうち、ペルオキシソーム局在型SPTの合成を担う下流側開始部位(+66)を支配するプロモーター領域は+21〜+90と同定された。HepG2細胞を用いたゲルシフト解析およびCATリポーター解析(変異導入解析を含む)の結果、この領域内に存在する一カ所のinverted repeat配列と、三カ所のC/EBP配列が下流プロモーター活性に寄与していることが示された。inverted repeat配列に結合するタンパク因子は同定できていないが、C/EBP配列にはC/EBPα及びC/EBPβが結合する。そこでC/EBPαまたはC/EBPβの過剰発現を行うと、どちらもCATリポーターの活性を顕著に増強し、この効果はC/EBP特異的ドミナントネガティブ体の共発現で阻害された。ミトコンドリア型SPTの合成を担う上流側転写開始部位(+1)のみを持つSPT遺伝子断片を用いたルシフェラーゼリポーター解析からは、-125〜-89と-14〜+10の領域が上流側プロモーター活性に寄与することが分かった。-14〜+10内にはAP-2認識部位様配列が存在する。この領域への核タンパク質結合は抗AP-2β抗体の共存による阻害が顕著であった。またAP-2βの過剰発現により、上流プロモーターのbasal活性のみが著しい上昇効果を受けた。一方、-125〜-89内のSp1認識部位様配列にはSp1が結合することが分かった。この部位の変異によりその結合阻害と上流プロモーター活性の低下が観察される。またDNA結合領域のみから成るSp1欠失体の過剰発現によっても活性の低下がみられる。野性型Sp1の過剰発現は上流プロモーターのbasalとPKA(protein kinase A)-induced活性の両方を上昇させ、この効果は転写コアクチベーターCBPの共発現によりさらに強められた。CBPの関与はadenovirusE1Aの共発現により阻害を受けることからも確かめられた。
