微生物感染・炎症における遺伝子損傷の分子メカニズム

研究概要	本研究では、微生物感染に伴う炎症局所において産生する活性酸素(スーパーオキサイドや過酸化水素)や窒素酸化物(一酸化窒素など)およびそれらの反応産物(パーオキシナイトライトなど)によりもたらされる遺伝子損傷を明らかにし、炎症から発癌へと続く分子メカニズムにおける活性酸素、窒素酸化物の役割を検討する。昨年度は、パーオキシナイトライトがヌクレオシド、RNA、DNAのグアニンを効率良くニトロ化し、ニトログアニン(ニトログアノシン)を生成することを報告した。また、生成したニトログアノシンがシトクローム還元酵素の存在下に顕著なスーパーオキサイドを産生することを明らかにした。本年度はさらに、このようなスーパーオキサイドの生成が細胞内で起こるのかを蛍光指示薬である2,7-ジクロロフルオレシンを用いたフローサイトメーターにより検討した。その結果、培養細胞をニトログアノシンとともに培養すると、濃度依存的に細胞内にオキシダントの生成が見られた。以上の結果は、パーオキシナイトライトによるグアニンニトロ化が細胞内で新たな活性酸素の生成をもたらし、細胞の酸化ストレスを高めることを示唆しており、パーオキシナイトライトによる遺伝子変異作用を解明する上で重要な知見を与えると考えられる。
