研究概要

腸炎ビブリオの2つの環状レプリコンよりなる染色体について以下の研究をおこなった。
1.1つの細菌細胞内で2つの染色体の複製がどのように制御されているかについて理解するために、腸炎ビブリオの染色体の複製起点の同定を行っている。現在までに大きい方の染色体における複製起点の位置が特定されている。
2.腸炎ビブリオの2つの染色体が正しく娘細胞へ分配される機構について解析を行うため、細菌細胞内での染色体の動態をFISH法により解析を行う実験系のセットアップを行っている。
3.ビブリオ属菌がいかにして2つの染色体をもつに至ったかについて、ゲノム構造を解析することにより考察を行っている。その手始めとしてビブリオ属に属するさまざまな菌種についてintactゲノムのパルスフィード電気泳動による解析を行った。その結果、すでにわれわれが報告していたコレラ菌やVibrio unlnificus,V.fluvialis以外にも多くのビブリオ属細菌で2つの染色体よりなるゲノム構造がみられ、このゲノム構造がビブリオ属菌間に広く保持されているものであることが明らかになった。
以上のような成果を踏まえ、残る1年間に以下の目標を達成する。
1.腸炎ビブリオのもつ2つの染色体の複製起点を同定する。
2.腸炎ビブリオの2つの複製起点および既知遺伝子をプローブとしたFISH法を行い、細胞分裂時における2つの染色体の動態を明らかにする。
3.さまざまなビブリオ属菌のゲノム構造に関して得られた情報をもとに、2つの染色体からなるゲノム構造が系統進化上いつ頃形成されたかについて考察する。
