エイズウイルスの増殖抑制に関与するCD8陽性T細胞由来因子の研究

研究概要

HIV-1感染無症候キャリアー(AC)の末梢血CD8陽性T細胞はHIV-1抑制活性を有することが知られており、この抑制には複数の機序が関与していると考えられている。我々はこれまでにHIV-1抑制機序と病態の進行との関連について検討を行い,AC期においてMHC-I拘束性CTL活性と非拘束性抑制活性が保たれている一方で、AIDSへの移行期において後者が先行して減弱する可能性を示してきた。本研究においては、このMHC-I非拘束性抑制活性に関与する因子の同定、および詳細な作用機序を解明するために、HIV-1の複製を容易に検出・定量できる新たな定量系を確立し,それを用いてCD8陽性T細胞による抑制活性の検出を試みた。
Green Fluorescent Protein(GFP)遺伝子がHIV-1遺伝子に組み込まれ、しかも複製可能なウイルス(HIV-1.GFP)は、蛍光強度を指標にして簡便にウイルス定量が可能であることが報告されている。そこでヒトT細胞株であるkit225細胞にHIV-1.GFPを感染させ、HIV-1.GFP慢性感染細胞株の樹立を試みた。得られた慢性感染細胞株(kit225/HIV-1.GFP)は蛍光顕微鏡により100%の細胞で蛍光発色が確認され、またFACSを用いた解析で単一の蛍光ピークを持つことが示された。さらにこの細胞とAC由来CD8陽性T細胞を共培養したところ、調べた中の約2/3のCD8陽性T細胞において蛍光強度を減弱させる活性、すなわちHIV-1抑制活性が確認された。本研究により樹立した細胞株はCD8陽性T細胞によるHIV-1陽性活性の詳細な解析に有用であると思われる。
