研究概要	インターロイキン(IL)-15は、1994年に新しいT細胞増殖因子として報告された。そのシグナル伝達にはIL-2レセプターbeta鎖およびgamma鎖を用い、IL-2と類似した生物活性を示す。しかし、IL-15の産生細胞はIL-2と異なり、上皮細胞、マクロファージなど、広い組織に分布している。実際、様々な感染症においてIL-15が上皮細胞やマクロファージから産生されることが報告されている。このことから、IL-15は局所における感染症や腫瘍に対する防御に深く関与するものと考えられる。我々は、IL-15の産生調節機構を調べる過程で、選択的スプライシングによって少なくとも2種類のmRNAアイソフォームが存在することを見い出した。従来のIL-15mRNAがコードするタンパク質は分泌型シグナルペプチドを持つが、新たに見い出したアイソフィソフォームは、シグナルペプチドを持たないタンパク質をコードするため、細胞外へ産出されない。アイソフォームの役割を調べるため、それぞれのIL-15cDNAをH-2Kプロモーター制御下で発現するトランスジェニック(tg)マウスを作成した。非分泌型IL-15tgマウスでは、内因性のIL-15タンパク質の産生が抑制され、細胞内寄生細菌に対して易感染性を示した。一方、分泌型IL-15tgマウスでは、NK、NKT細胞、メモリータイプCD8T細胞および子宮上皮間gamma/delta型T細胞の著しい増加が認められ、細胞内寄生菌に対して抵抗性を示した。分泌型IL-15tgマウスで増加するメモリータイプCD8T細胞は、感染早期においてIL-12/IL-15によってバイスタンダーに活性化され大量のINF-gammaを産生し、感染防御に働くことが分かった。これらの研究から、IL-15mRNAの選択的スプライシングによって新たに生じたアイソフォームは、生体局所における過剰なIL-15の産生を制御している可能性が示唆された。今後、非分泌型IL-15tgマウスを用いて分子レベルでのIL-15産生制御に関する解析を行う予定である。
