冠攣縮の病因解明:変異型eNOS遺伝子(-786C)の転写を抑制するRPAの意義

研究概要	1.eNOS遺伝子の-786C変異とRPA1の発現がeNOS遺伝子発現に及ぼす影響の検討 RPA1(replication protein A1)はこれまで遺伝子の修復に関与する核蛋白質であると考えられていた。ウエスタン・ブロット法で検討したところ、臍帯静脈、大動脈、冠動脈それぞれ由来のヒト血管内皮細胞にてRPA1の蛋白発現が確認され、内皮型一酸化窒素合成酵素遺伝子(eNOS)遺伝子の野生型及び変異型プロモーターを繋いだルシフェレースレポーター遺伝子をトランスフェクションし、転写活性を比較したところ野生型プロモーターに比べ変異型プロモーターはその転写活性は約20%の低下を認めた。 2.生体組織におけるRPA1の発現と-786C変異がeNOS遺伝子発現に及ぼす影響の検討ヒトの生体組織においてRPA1が血管組織に発現し、-786C変異がeNOS遺伝子の遺伝子発現に影響を与えているかを、血管組織が豊富なヒト胎盤組織を用いて検討した。ウエスタン・ブロット法にて胎盤組織にRPA1が存在することが確認され、遺伝子型を確定した胎盤組織において-786変異を有する胎盤組織では、野生型ホモに比べeNOS遺伝子の発現が減少していることがRNase protection法にて確認された。また、eNOS蛋白質の産生物質であるNOの血清レベルをNOxを測定することで検討したところ-786変異を有するヒトは野生型ホモに比べ、そのNOxレベルが有意に低値であり、-786変異を有するヒトにeNOS蛋白の量的あるいは機能的低下が存在することが予想された。現在さらに、eNOS遺伝子の-786変異と動脈硬化性疾患及び、糖尿病やその血管合併症の成因との関連を検討している。
