重粒子線と温熱との併用療法の基礎的研究

研究概要	我々は難治癌に対する治療の向上のため、高LET線源に特徴的な修復不能なDNA損傷を引き起こす重粒子線に注目し、より効率良く癌細胞に致死をもたらすことを期待して、温熱処理との併用による基礎研究が重要と考えた。今回、我々はp53遺伝子を導入した神経膠芽腫細胞における重粒子線のLETに対する殺細胞効果を明らかにし、さらに温熱との併用におけるp53依存性およびLET依存性について検討した。その結果、低LET放射線による殺細胞効果にp53遺伝子型が寄与していることが示唆された。このことは、あらかじめ患者の癌細胞について正常型p53の機能を持ち備えているかどうかを調べることが、低LET放射線による癌治療方針の決定に重要であることが示唆された。また、炭素線がX線に比べて高い殺細胞効果を確認した。重粒子線療法はp53遺伝子型によらず治療効果が期待できることが示唆された。特に、悪性腫瘍の半数とも言われる変異型p53の癌細胞をもつ患者にも、重粒子による高LET放射線照射は、有効性が期待できる。さらに、正常型p53の癌細胞のみ、X線と同様に、現在治療で用いられているLET70 KeV/μmの炭素線の場合でも温熱との併用による相乗的な殺細胞効果を確認した。この結果から、難治癌に対する集学的療法の一つとして炭素線と温熱併用の適用が期待される。今後さらに、移植腫瘍での研究をすすめるとともに、放射線と温熱の相乗作用に及ぼすp53の役割についてのメカニズムを解明していきたい。
