研究概要	コレラ毒素(cholera toxin:CT)はタンパク抗原やワクチンに対する強い粘膜アジュバント作用を有するが、ADP-ribosyltransferase活性による下痢誘導作用によりヒトに応用できない。申請者らは米国アラバマ大学ワクチンセンター,国立国際医療センターと共同で無毒化CT(mutant CT:mCT)を作製し,これらのmCTが下痢誘導作用を持たずに粘膜アジュバント効果のみを有することを示した。本研究ではCD4^+ T細胞あるいは抗原提示細胞による免疫応答誘導においてmCTがどのような効果を及ぼしているのかを両免疫担当細胞間のネットワークを含めて細胞と分子のレベルで解析を加えた。その結果、mCTはパイエル板および脾臓のB細胞、マクロファージに作用してB7-2を誘導した。さらにmCTによって活性化されたこれらの抗原提示細胞はTCR-CD3複合体を介して刺激を加えられているCD4^+ T細胞の細胞増殖を促進した。また、その誘導活性は野生型(native CT:nCT)と同程度であった。一方、mCTはCD4^+ T細胞に直接作用することにより、Th1型サイトカイン産生を選択的に抑制した。同様の実験をCT-Bサブユニットを用いて行ったが、CT-BサブユニットにはB7-2の誘導能やTh1型サイトカインに対する抑制効果は認められなかった。これらの結果から、CTのアジュバント効果はAサブユニットに依存しているが、毒素活性は必要ではないことが示された。
