シスイソプレン鎖延長酵素の構造と触媒機構の解明

研究概要

Micrococcus luteus B-P26のウンデカプレニル二リン酸合成酵素を大腸菌中に大量発現させる系を構築し、酵素を大量に精製して、結晶化することに成功した。X線回折による高次構造解析を行い、シスイソプレン鎖延長酵素として初めてX線結晶構造を解析した。各サブユニットは10本のα-ヘリックスと6本のβ-シートから成っており、トランス鎖延長酵素と全く違う立体構造を持っていることが分かった。また、シスイソプレン鎖延長酵素に保存されている5つの領域はこの酵素の疏水性キャビティーの周りに存在することが判明した。
また、部位特異的変異導入によるウンデカプレニル二リン酸合成酵素の触媒機能発現機構の解明を行った。以下の知見が得られた。
1.77位Asnはアリル性基質の二リン酸部分に作用し、直接に触媒機能に関与している。78位のTrpはアリル性基質の疏水性プレニル鎖の結合に参与する。
2.Region VにあるArg-197、Arg-203、Glu-216はホモアリル性基質の二リン酸を認識し、基質の結合や触媒機能に関与する。このことにより、シスイソプレン鎖延長酵素の基質認識機構はトランス鎖延長酵素と全く違うことが分かった。また、Region IIIのFS motifは同じ基質の疎水性炭素鎖及び二リン酸の結合に参与する。
3.Region Iにある"Structural P-loop"、Asp-29、Arg-42はアリル性基質の二リン酸の結合に必須である。
4.Phe-73、Ser-74及びGlu-216は生成物のプレニル鎖長の決定に参与する。
現在、アリル性基質のプレニル鎖部分の結合部位の解析を部位特異的変異導入及びこの酵素のPhotoaffinity labelingにより行っている。
