研究概要	プロテインCインヒビター(PCI)は、血液凝固制御因子の活性化プロテインC(APC)に特異的な血漿阻害因子で、α1アンチトリプシンなどと相同な構造を有する血漿セリンプロテアーゼインヒビター(SERPIN)ファミリー蛋白質の一つである。また、PCIは精奬中に血漿中の約40倍も高濃度に存在し、男性不妊症患者精奬中では著しく低下していること、また精子の先体にも存在し、卵との受精に関わっていることが示唆されいる。申請者は、これまでに本奨励研究の補助の基、肝癌由来株HepG2細胞でのPCIの発現とその制御に重要な転写調節領域を解析し、PCI遺伝子の肝における発現のプロモーター領域としてはSp1結合部位が、エンハンサー領域としてはAP2結合部位が重要で、これらの領域にはHepG2細胞由来のSp1およびAP2蛋白質がそれぞれ結合すること、またサイレンサー領域としてはA-activator結合部位およびインターフェロン-γ反応領域が重要で、これらの領域にはHepG2細胞由来の未知の核蛋白質が結合することを報告してきた。同時に、PCIが血小板・巨核球でも発現され、血小板リン脂質表面上でAPCを効果的に阻害すること、およびラットではPCIが男性生殖臓器(精巣や精嚢腺)で特異的に発現し、その発現がアンドロゲン依存症であることを報告した。本年度は、これまでの研究をさらに発展させ、ウシPCIcDNAのクローニングを行い、そのアミノ酸構造および組織分布を明らかにすると共に、そのプロテアーゼ阻害特性に関して検討を行った。その結果、ウシPCIは主として肝臓、腎臓および男性生殖臓器で発現され、その反応部位は、ヒト、サル、マウスおよびラットPCIではArg-Serであるのに対して、ウシPCIではLys-Serであること、およびヒトPCIがヒトプラスミンを阻害しないのに対して、ウシPCIがウシプラスミンを一過性に阻害することが明らかになった。以上の検討から、ウシPCIはウシの血液凝固系だけでなく線溶系でも機能することが示唆された。
