研究概要	本研究で明らかになったことは、主に次の3つである。1.[α-ラクトアルブミン(α-LA)のモルテン・グロビュール(MG)状態の構造および安定性の解析]ヒトα-LAのMG状態及び巻き戻り中間体の構造と安定性を測定した結果、ヒトα-LAのMG状態は他のα-LAのMG状態よりも安定であり、かつ、より多くのα-ヘリックスを含むことが明らかになった。 2.[α-LAの変異体のフォールディング反応の解析]α-LAの様々な変異体を作成するために、ヤギα-LAの遺伝子をpSCREEN-1b(+)に組み込み、蛋白質の発現を行った。変異体を使った研究から、α-LAのフォールディング反応の遷移状態では、T29,I95,W118周辺の構造はまだ十分には形成されていないことが明らかになった。このことは、MG状態で形成される疎水性コアやCヘリックス周辺は遷移状態において構造化されていないことを示している。 3.[ストップトフローX線溶液散乱法によるα-LAの巻き戻り反応の測定]従来のストップトフローX線溶液散乱法では一次元PSPC型X線検出器を用いて測定を行っていたが、新たに開発された二次元CCD型X線検出器を用いることにより、400倍以上の感度向上に成功した。また、データの補正方法を確立した。本研究で完成した時分割X線溶液散乱法は、蛋白質のフォールディング研究のみならず、様々な研究に適用可能な優れた方法である。この方法を用いてα-LAの巻き戻り反応の測定を行った結果、α-LAは巻き戻り反応開始後数10ミリ秒以内に、平衡条件下で観測されるMG状態と同じ分子サイズと分子形状を持つ巻き戻り中間体を形成することが明らかになった。本研究の結果およびストップトフロー円二色性法による結果から、α-LAの巻き戻り中間体は平衡条件下で観測されるMG状態と同一であることが示された。
