FTIRによる光合成酸素発生マンガンクラスターの構造の解明

研究概要

閃光誘起フーリエ変換赤外(FTIR)法を用いて、植物の酸素発生反応を担う酸素発生系(光化学系II蛋白質複合体の一部として存在)の構造と反応を調べた。まず、基質である水分子のFTIRバンドを検出し、その構造の知見を得た。暗順応した酸素発生系のS_1状態から、250Kでの一閃光照射によって得られるS_2中間状態への遷移の際のFTIR差スペクトルを測定すると、3618/3585cm^<-1>に微分形のバンドが観測された。H_2^<18>O置換した試料で測定することにより、これは酸素発生系中の水分子のO-H伸縮振動であると同定された。さらに、D_2Oによるデカップリング実験から、観測された水分子は非対称的な水素結合状態を形成しており、S_1からS_2状態への遷移により、その非対称性が増すことが明らかとなった。また、常温において1s間隔で連続的に閃光照射を行い、その間のスペクトルを順次測定することにより、酸素発生系の反応サイクルにおけるすべての閃光誘起遷移(S_1→S_2,S_2→S_3,S_3→S_0,S_0→S_1)のFTIR差スペクトルを測定することができた。得られたスペクトルから、S_1→S_2→S_3遷移におけるタンパク質の構造変化(カルボキシル側鎖やタンパク質二次構造の変化など)が、S_3→S_0→S_1遷移において反転する様子が観測された。
