逆遺伝学的手法による高等植物シグナル伝達経路関連遺伝子の機能解析

研究概要

シロイヌナズナの5番染色体のctr1遺伝子座の近傍にDsT-DNAが挿入された親植物系統(Ds388-30)を用いてDsを転移させ、これまでに転移植物系統を約1200系統作製した。作製した系統についてDsの挿入により破壊された遺伝子断片をTAIL-PCR法を用いて回収し塩基配列決定後、相同性検索を行ったところ、乾燥ストレスにより誘導される遺伝子でAquaporin遺伝子のRD28遺伝子やオーキシンのシグナル伝達に関与するAUX1遺伝子のホモログ遺伝子などの遺伝子内部にDsが挿入した系統が存在していたので、本年度はまず、それら系統についてDsの挿入変異をホモにもつ個体を作製し、表現型を調べた。しかしながら、野生株と比較して表現型の異常は見い出されなかった。
また、これまでに作製した系統の内約500系統について、Dsの挿入変異をホモにもつ個体を作製し表現型を調べた。その結果、アルビノの表現型を示す系統が2系統(geranylgeranyl pyrophosphate synthaseのホモログ遺伝子の破壊株、TATC遺伝子の破壊株)存在していた。これら系統についてはDsを転移させ変異が回復することの確認を行う予定である。
また、新たに作製した系統について引き続きDsの挿入により破壊された遺伝子断片の塩基配列決定および相同性検索を行った。その結果、ソラマメのABA-activated Ser/Thr protein kinaseのホモログ遺伝子、Phosphatidylinositol 4-kinase遺伝子、Expansin遺伝子(At-Exp2)などの遺伝子内部にDsが挿入した系統が作製されていることがわかった。
