WASPファミリー蛋白によるアクチン細胞骨格制御機構の解明

研究概要

まず、N-WASPがアクチン重合を誘導するメカニズムとして、アクチン重合核形成誘導因子Arp2/3complexを活性化することを明らかにした。N-WASPのC末部にはアクチンを制御するVCA領域があるが、そのうち最もC末部に位置するCA領域がArp2/3complexに、そしてV領域が単量体のアクチンとそれぞれ直接に結合した。このとき、Arp2/3complexの持つアクチン重合核形成能が極めて顕著に上昇し、in vitroでのアクチン重合のスピードを促進した。このVCA領域はWASPや後述するWAVEにも存在しており、WASPファミリー蛋白はArp2/3complexの制御因子群であることが示唆された。さらに全長のN-WASP蛋白ではこのVCA領域がマスクされており、普段はArp2/3complexを活性化できないが、Cdc42やPIP2の添加によりそれが解除されVCA領域単独のものと同程度の活性が観察され、N-WASPの機能制御に関わる重要な知見を得た。
次に他のWASPファミリー蛋白としてデータベース中に機能不明のcDNAクローンを見つけ、それをWAVEと名付けた。この他にも、そのWAVEとよく似た蛋白を更に2つ見つけ出しそれらをまとめてWAVE1、2、3、と命名した。これらWAVEsはWASPやN-WASP同様Arp2/3complexを活性化するVCA領域を持っていたが、Cdc42との結合領域はなかった。実際、変異体発現による解析からWAVEsはCdc42の下流でなくRac下流において機能し、ラッフル膜形成に関与する蛋白であることを明らかにした。
