生体膜リン脂質の非対称構造を規定する分子機構の解明-細胞遺伝学的アプローチ-

研究概要	私は、生体膜上のリン脂質であるホスファチジルエタノールアミン(PE)に対して特異的に結合するペプチドを用いて、様々な細胞活動に伴うPEの膜動態について解析を行なった結果、細胞分裂期において、通常は形質膜内層に局在しているPEが一過的かつ分裂溝(細胞がくびれる部分)特異的に表層に現れることが明らかにしている。リン脂質は親水性の頭部を有するために、疎水性の二重膜を横切る運動(フリップ-フロップ運動)を自発的に行なうことは困難である。従って、分裂期にPEが一過的に形質膜外層に現れ、それが分裂終了後に速やかに内層に戻るためにはPEを積極的に動かす分子機構の存在が想定される。私はPE輸送に介在する分子機構を明らかにするために、変異剤処理した酵母細胞5万株の中からPE結合ペプチドに対する感受性が異常になった変異株17株を樹立した。その中の1つros3株は、形質膜上におけるPEのフリップ-フロップ速度が親株と比較して顕著に低下していた。PE結合ペプチドに対する感受性を指標にしてros3株の形質を相補する遺伝子を単離したところ、2回膜貫通領域を有すると予想される機能未知のタンパク質(Ros3p)をコードしていた。ポリクローナル抗体を作成し、その細胞内分布を検討したところ、Ros3pは主として形質膜に分布した。次に、ros3遺伝子を相同組み替え法により特異的に欠損させた株を作成し、その性状を解析したところ、細胞増殖には異常は見られないが、出芽の方向性、アクチンパッチの重合に異常が見られた。さらにGal7プロモーターを用いてros3遺伝子を過剰発現させると1つの細胞から多数の出芽が起きるという出芽異常が観察された。以上の結果から、Ros3pが形質膜上でPEの配向性を変化させることによって、アクチン骨格の重合位置、さらには出芽の方向性を制御している可能性が考えられた。
