ショウジョウバエの発生におけるRas情報伝達系の機能と制御機構

研究概要

ショウジョウバエEGF受容体を介するシグナル伝達経路について解析を行い以下の成果を得た。
(1)EGFモチーフを有する分泌性蛋白質Argosを複眼特異的に過剰発現するトランスジェニック系統GMR-argosのtenetic modifierとして分離した変異の染色体上における位置を決定し、近傍に位置する既知の遺伝子の変異体との相補性試験を行った。分離した変異体の一部は既知のEGF受容体シグナルに関与する遺伝子の変異であり、残りは新規な遺伝子の変異と考えられた。
(2)バキュロウイルス発現系を用いて得たAgros、Spitz(ショウジョウバエのTGFα相同分子)及びEGF受容体の蛋白質を用いて、これらの結合活性を調べた。BIAcoreおよび免疫沈降法により、AtgosとSpitzがいずれもEGF受容体の細胞外ドメインに結合することが明らかになった。また、ウエスタンブロッティング法によりArgosはEGF受容体のニ量体形成を直接阻害することが明らかになった。
(3)Drosophila Ralのドミナントネガティブ及び恒常的活性型変異体のショウジョウバエ個体での強制発現実験により、Ralが発生過程における細胞形態の変化の制御に関与する可能性が示唆された。遺伝学的交配実験及び培養細胞への強制発現とリン酸化JNKの特異抗体を用いたウエスタンプロッティングによって、RalにはJNK経路を負に制御する活性があることを明らかにした。
