組織特異的幹細胞を用いたヒト人工組織生産技術の開発

研究概要	正常ラット肝臓から幹様細胞を分離し、培養・維持することができた。この幹様細胞をコラーゲン内の3次元培養系で蛋白質の変化を調べたところ、アルブミンを発現していなかった幹様細胞が1週間後からアルブミンを発現し始め、徐々に発現量を増加した。以上の結果、肝臓幹様細胞を3次元培養することによって、アルブミンを発現する、より成熟した細胞に分化させることができた。次に、毛髪再生に関する成果として、ヒト皮膚の幹細胞に特異的と知られているサイトケラチン(CK)15が、マウスの毛髪組織でも毛幹細胞のマーカーになりうることを発見した。この成果は、今後エレクトロポレーションで蛍光標識用遺伝子(GFP)を導入した毛幹細胞をCK15をマーカーにして分離できる目途がついた。骨組織再生に関する成果として、軟骨への分化を誘導する遺伝子を探究するため、ヒト胎児軟骨細胞培養系からサブトラクション法で17個の未知遺伝子断片をクローニングした。そのうちDEC1は新規のプロリン型bHLH転写因子であり、CDEPは新規のRhoGEFであった。また軟骨に特異的に発現する膜蛋白MTFをクローニングした。ATDC5細胞にDEC1あるいはMTFを過剰発現させると軟骨分化が亢進した。一方3T3細胞へCDEPを導入すると細胞が癌化した。自家軟骨細胞あるいは骨髄間葉系細胞の移植による軟骨/骨の再建法に、DEC1とMTF遺伝子は有用な道具となると期待される。
