高度好熱菌を使用した廃水処理に関する研究

研究概要	九州地区において、好熱菌を利用した廃液処理を開発を目的としている。研究内容は二種あり、第一は、九州地区の地熱発電所における硫化イオンを含んだ廃液を、超好熱性好酸性古細菌Subfolobus sp. Strain 7を用いて硫化イオンを硫酸イオンにまで酸化して安全に廃棄できるようにするプロジェクトである。廃水処理技術としては、試験段階ではほぼ完成に近づいているが、基礎研究が遅れているので、本研究計画では本菌の呼吸系の解明を行っている。この菌には、Zuを含むという珍しいタイプのフェレドキシンが存在するが、このフェレドキシンの構造とZnの役割について研究を進めた。また、呼吸系の複合体を分離、各コンポーネントを単離して、それらをコードしている遺伝子を明らかにし、再構成を試みる研究が進展した。この研究は三菱重工長崎造船所、同横浜研究所との共同研究である。もう一つのプロジェクトは、鹿児島市の下水処理場の汚水コンポスト化に関するもので、株式会社「山有」との共同研究である。本研究においてはコンポスト化に関わる好熱性細菌の単離同定を目的として、同社が提供する堆肥試料から好熱菌の単離を行った。その結果、中等度好熱菌の単離に成功した。そのリボソームRNAの塩基配列を解析し、系統解析を行った結果、従来のコンポスト化に関わるBacillus stearothemophilusとは異なる種の細菌であると結論された。さらに、他の好熱菌の存在も明らかとなっているので、単離を試みることを継続しているが、継代培養には成功していない。
