犬のアレルギー疾患におけるin vitro診断法の確立

研究概要	最近の疫学調査でアレルギー疾患の犬の20%程度がスギ花粉アレルゲンに感作されていることが明らかになってきた。これらのイヌはスギ花粉アレルゲンに対して皮膚反応やIgE抗体陽性反応を示す。しかしながら、これらのイヌにおいて、アレルゲン検査法として、スギ花粉主要アレルゲンに対するヒスタミン遊離やT細胞の反応性は調べられていなかった。今研究においてアトピー性皮膚炎の犬におけるスギ花粉アレルゲンに対するヒスタミン遊離やT細胞の反応性を調べた。スギ花粉粗抗原特異的IgE抗体陽性のアトピー性皮膚炎の症状を示すイヌ10例の中、6例について末梢血リンパ球を分離して、スギ花粉アレルゲンで刺激後、H^3-thymidineの取り込みで調べた。6例中、5例においてスギ花粉アレルゲンに対するT細胞増殖が認められた。この結果により、スギ花粉アレルゲンに感作されたイヌにおいて、スギ花粉アレルゲン特異的細胞性反応が初めて認められた。in vitro法のアレルゲン検査法として、アレルゲン特異的細胞性反応は有用であると考えられる。次に上記の10例中、6例において末梢血中の好塩基球からのスギ花粉アレルゲン特異的ヒスタミン遊離を調べた。6例中、3例は比較的多くの好塩基球が回収できたが、他の3例は0.1%以下であった。6例中、2例にスギ花粉特異的ヒスタミン遊離が認められたが、他の4例は自然遊離と変わらなかった。遊離反応が測定できなかったイヌにおいては末梢血の好塩基球が少ないため、特異的なヒスタミン遊離が認められなかったと考えられた。今後、イヌにおいてアレルゲン特異的ヒスタミン遊離をin vitro法のアレルゲン検査法として確立していくには、もうすこし工夫が必要であると考えられた。
