染色体バンド境界の分子レベルでの特定とその機能探索

研究概要	高等脊椎動物の染色体バンド領域は、S期内の複製時期やGC%のMbレベルの区分構造に関係しており、遺伝子密度や発現様式を異にしたゲノムの機能ドメインと考えられるようになってきた。バンド境界は複製タイミングのスイッチ点やGC含量のモザイク境界として、塩基配列レベルで特定できる可能性が高く、境界を特徴づけるシグナル群が存在する可能性が考えられる。ヒト21番染色体長腕の全域と、ヒト11番染色体の長腕の全域を対象に、詳細な複製時期地図をゲノム配列レベルで作成した。染色体11qおよび21qの両方において、S期前半と後半の複製領域がRとGバンド領域に対応することが判明し、複製時期がS期前半から後半に明瞭に転換している領域を約30箇所、塩基配列レベルで特定できた。GC含量の区分境界と複製時期の転換部位とが密接に関係することも明らかになり、これらの転換部位をバンド境界の構造上ならびに機能上の要件を満たす部位として塩基配列レベルで特定した。バンド境界に対応する複製時期の転換部位には病因遺伝子、特に脳神経疾患遺伝子および癌関係遺伝子が集中して存在するとの予想外の知見を得た。複製時期の転換領域では、DNA複製フォークの進行が休止する可能性が指摘されており、組み換え頻度が上昇していると予想されてきた。染色体転座や遺伝子増幅と関係した癌関係遺伝子が、この領域に局在することと関係すると考えられる。バンド領域は複製時期と関係するだけではなく、クロマチン構造の粗密度と関係することも知られている。従って、染色体バンド境界においてはクロマチン構造の粗密度が転換している可能性が考えられる。バンド境界の持つこれらの特殊事情が、病因と関係する可能性を考えている。今回の研究は、病因の分子機構を知る手がかりを与えるだけでなく、あらたな病因遺伝子を能率的に探索する方法としても発展すると予想している。
