深海由来の高圧遺伝子群の開発に関する研究

研究概要	本研究の目的は深海ならびに深海底以深の岩盤層に生息している微生物の分離ならびにそれらがもつ遺伝子を資源として活用することにある。そのためには、種々の深海環境から海水、底泥、岩石、あるいは熱水孔等を採集し、濃縮した後にDNAを抽出し、ライブラリーを作成するところから始まる。 11年度では、太平洋南西諸島海域、トカラ列島島小宝、長崎県橘湾、京都大学植物園等から得た試料につき、DIFCO nutrient brothを用いて高圧条件下(40Mpa)、10度Cならびに30度Cで集積培養をおこなった。いずれの試料からも、この条件下で増殖してくる細菌が存在し、培地を白濁させた。このことは、単に深海といった常に高圧条件にある環境以外の一般的には高圧条件下にない環境にも耐圧性の細菌が存在することを示している。現在、このように高圧条件下で増殖してきた細菌を、限界希釈法ならびに塗抹法を用いて分離するとともに、DNAの抽出とライブラリーの作成、16S rRNA遺伝子情報に基づく系統解析が進行中であるが、真正細菌以外に古細菌の仲間も存在している可能性が示唆されている。また、高圧条件は高温条件のアナロジーととらえて、分離されてくる微生物と好熱細菌との類似性についても注目して研究を進めた。この研究費で購入した、高圧培養大型容器は、大量の試料を高圧条件で処理できるので研究に活用することができた。
