磁力は骨芽細胞の分化・増殖を著しく亢進させる-その分子機構解明と再生医学応用-

研究概要	研究代表者は動物細胞への遺伝子導入効率を上げる目的で、遺伝子の運搬役として微小磁性体(magnetic microparticle;0.05mm)の使用を思いつき、それをエレクトロポレーションによりマウス頭蓋冠骨芽細胞(Osteoblast;MC3T3-E1株)に導入することに成功した。その細胞に永久磁石(希土類磁石)で細胞の水平方向に正磁場をかけたところ、骨芽細胞(骨細胞に分化する)が両極方向に伸展した。その際、対照として、(1)磁性体を導入しないで磁場だけをかけたもの、(2)磁場をかけず、磁性体だけを導入しただけのもの、(3)磁性体も導入せず、磁場もかけないもの等について、細胞の増殖・分化度を顕微鏡レベルで解析した。その結果、磁性体を導入し、かつ、磁場をかけた骨芽細胞の培養を継続すると、細胞の結節形成が早まり、その周囲には燐酸カルシウムと推定される結晶がきわめて多く観察された。このことは、メカニカルストレッチあるいは磁力の影響により、ある遺伝子のスイツチがONとなり、それが引き金となって細胞の増殖や分化を誘導したことを示す。そこで、次に、同様な実験を正常ヒト骨芽細胞(NHOst)細胞を用いて試みた。先ず、NHOst細胞の培養条件を検討した。その結果、最適培地においてコンフルエントまで6〜9日ほどかかることが分かった。また、MC3T3-E1細胞ではハイドロキシアパタイトが凝集した結晶が認められるのに対し、NHOst細胞ではvonkossa染色で石灰化が確認されるのみであった。
