研究概要

1.B95aマーモセットB cell上のヒトCD46 homologueの同定
マーモセットB95a細胞株から2種のCD46様分子をcDNAクローニングした。1つは4つのshort consensus repeat(SCR)、1つは3つのSCRを含む細胞外ドメインを持つ。N末1個のSCR(SCR1)の有無からmRNAのalternative splicingにより2種のspliced formが生じることが判明した。SCR1^+isoformが90%、SCR1^-isoformが10%の比で膜上に存在し、SCR1^+isoformのみが麻疹ウィルスワクチン株CAMのレセプターとして機能した。他のウィルス株はwild-typeを含めてマーモセットCD46をレセプターとして使用しなかった。B95aの麻疹ウィルス高感受性はCD46以外のレセプター分子に起因する。
2.B95a上の麻疹ウィルスセカンドレセプターの同定
B95aをマウス(Bal b/c)にくり返し腹腔(i.p)又は皮内(s.c)投与し、脾細胞からhybridomaを樹立した。Nagahata株、最近のwild-type strain(Yokoyama株)の合胞体形成阻害活性をクライテリアにセカンドレセプターの阻害抗体をスクリーニングした。6回Fusionを試行し、計3,500個以上のhybridomaをスクリーニングしたが完全阻害抗体は得られなかった。部分阻害抗体は4株得られ、うち2種は分子量約2万の同一分子を認識した。他の2種はwestern blotで分子同定ができなかった。当該分子の同定を推進中である。
