肝発癌制御ワクチンの開発

研究概要

C型肝炎ウイルス(HCV)・コア遺伝子をもつトランスジェニックマウスを樹立した。このトランスジェニックマウスでは、早期に肝の脂肪化を起こし、後期に肝細胞癌を発生した。免疫惹起により肝発癌の抑制を図るため、コア蛋白をコードするDNAワクチンを投与した。通常、トランスジェニックマウスは発現蛋白に対して寛容状態となっているため、通常の抗原刺激では免疫反応が得られないことが多い。インターフェロン・ガンマなどのサイトカイン遺伝子を同時に発現させることによって、これを克服(寛容状態をブレーク)することが可能である。遺伝子型1bのHCVcDNAクローンよりコア遺伝子を切り出し、肝特異的発現ベクターpBEPに挿入し、DNAワクチンのコンストラクトを作製した。同一の制御領域のもとにサイトカイン遺伝子であるインターフェロン・ガンマ遺伝子を接続したコンストラクトを同様に作製した。これらのコンストラクトを肝癌由来培養細胞HepG2にトランスフェクトし、その蛋白発現、活性の確認を行なった。このコンストラクトを、HCVコア遺伝子トランスジェニックマウスの筋肉内に都合三回投与した。約1/3のマウスにおいてコア蛋白特異的抗体の産生が認められた。炎症細胞の浸潤等の炎症所見は認められていない。
