拒絶反応におけるIL-18の役割について

研究概要	当該研究は、移植医療に必然に併発する拒絶反応がIL-12とIL-18によって惹起・増幅される炎症病態であることを、これらのサイトカインが欠損した動物を用いて証明するものである。C57BL/6をレシピエントとしてMHC class Iの全く異なるBALB/cの皮膚片を移植すると、あるいは逆にBALB/cをレシピエントとして、C57BL/6の皮膚片を移植すると強い拒絶反応がおこる。我々はC57BL/6バックグランドのIL-18欠損マウスを既に作製している。そこで、このC57BL/6バックグラウンドのIL-18欠損マウスに野生型BALB/cマウスの皮膚片を移植し生着を経時的に観察した。野生型に比べてIL-18欠損マウスへの皮膚移植片の生着率はあきらかに良好であった。しかし、逆に、野生型BALB/cマウスにIL-18欠損或いは野生型C57BL/6マウス皮膚片を移植しても、両者の移植片生着率に差は認められなかった。これらの結果から、皮膚移植片に対する細胞障害にはIL-18が重要な役割を果たしていることが示唆された。現在、その詳細なIL-18の作用機序を解析中である。また、もし宿主の血中で、実際宿主由来のIL-18値が上昇していた場合、抗IL-18抗体を投与することで、拒絶反応がどの程度抑制されるかを調べる予定にしている。更に、C57BL/6バックグラウンドのIL-12/IL-18のダブルノックアウトマウス並びにIL-12単独ノックアウトマウスを用いて同様の検討をおこなうことで、拒絶反応におけるIL-12とIL-18の役割を解明したいと考えている。
