肝細胞癌に対するMAGE-1遺伝子産物ターゲットとした特異的免疫療法

研究概要

【背景・目的】
われわれは,肝細胞癌に高率(80%)にMAGE-1遺伝子の発現がみられることを報告した.本研究の目的は,MAGE遺伝子産物をターゲットとした肝細胞癌特異的な免疫療法の臨床応用が可能であるかどうか検討するものである.
【方法・結果】
肝切除術が施行された原発性肝癌症例を対象とし,腫瘍組織よりmRNAを抽出し,β-actinの発現を確認した後,異なるExon間にPrimerを設定し,RT-PCR法によりMAGE-1,2,3遺伝子の発現を観察した.また,肝臓由来の細胞株でも同様の検討を行った.肝癌組織で高率にMAGE-1および-3遺伝子の発現が確認された.切除肝あるいは生検肝を用い,免疫組織学的方法によりMAGE遺伝子産物の発現の検討を行っている.
抗原特異的免疫療法を行うためには,MAGE-1蛋白のT細胞エピトープを明らかにし,エピトープペプチドによるその反応性を検討することが重要である.MAGE-1蛋白のエピトープは、現在のところHLAA1でしか明らかにされていないため,別に研究を進めている原発性胆汁性肝硬変の抗原エピトープ特異的T細胞を用いて,自己抗原エピトープ・ペプチドのアナログペプチドに対するT細胞応答性を解析中である.
【今後の計画】
患者と同一のHLA class I抗原を有した細胞株にMAGE-1遺伝子を導入し,mRNAおよび蛋白レベルでのMAGE-1遺伝子の発現をRT-PCR法およびサザンブロッテイング法で確認する.さらに,患者から採取したTリンパ球を,HLA class I matched MAGE-1陽性細胞株をfeederとして培養し,^<51>Crリリース法を用い,feederとして用いた細胞に特異的な細胞障害活性を検討中である.
