レチノイン酸と、神経細胞死誘導性抗APP抗体を併用した神経芽細胞腫の萌芽的治療

研究概要	前年までの検討で、抗APPモノクローナル抗体はSH-SY5Y細胞に細胞死を誘導したが、NIE115細胞では細胞死は抗APP抗体によっては誘導されなかった。また、既に報告したように、NIE115細胞にレチノイン酸処理を行ってもAPP発現量の有意に変化は観察されなかった。しかし、レチノイン酸がAPP発現量を上昇させなかったからといって、一定量のAPP発現がNIE115細胞には見られる為、抗APPモノクローナル抗体がNIE115細胞に細胞死を誘導できないのは、APPが引き金をひく細胞死機構が欠除する為とは結論できなかった。そこでもう一度原点に立ち帰って考察した。NIE115細胞において抗APPモノクローナル抗体が細胞死を誘導し得ない理由には、2通りあり得た。第1が、抗体による細胞死誘導機構が不十分、もしくは存在しない場合。第2が、抗体による細胞死誘導機構は備わっているが、細胞死抑制機構が発現している為、最終的な細胞死が誘導されない場合である。これらを区別する為、SH-SY5Y細胞とNIE115細胞を融合し、抗APPモノクローナル抗体が細胞死を誘導するか否かを検討した。この目的の為、まず、SH-SY5Y細胞にPSV2neoを安定発現する細胞株とNIE115細胞にPBabe/puroを安定発現する細胞株を作成した。両者を融合し、ネオマイシンとピューロマイシンの両者に耐性の細胞株を選別した。しかし、得られた細胞株は増殖が遅く、最終的に継代培養可能な細胞株として樹立できなかった。現在、これに代わる方法として、SH-SY5Y細胞にEGFPを発現する細胞株と、NIE115細胞にdSRedを発現する細胞株を一過性に融合し、緑色蛍光と赤色蛍光とを同時に発生する細胞をFACSソーターにて分取し、分取された細胞群に対して抗APP抗体が細胞死を誘導するか否かを検討中である。
