婦人科腫瘍に対する電気穿孔法を用いた遺伝子治療のための基礎研究

研究概要	子宮平滑筋肉腫や卵巣腫瘍を含む婦人科悪性腫瘍において、個体レベルで悪性組織に抗癌剤や腫瘍抑制的に働く遺伝子を導入することによって、治療困難な悪性腫瘍の治療を行うための基礎的研究を行うことを目的として、本年度は簡便な治療法を確立するために遺伝子を使わず抗癌剤を電気穿孔法で悪性腫瘍組織に導入する治療法を検討した。子宮平滑筋肉腫細胞株SK-LMS-l細胞をヌードマウスに移植後、直径10mmの腫瘤が出来た段階で抗癌剤、特にブレオマイシンを導入した。電気穿孔法で導入する抗癌剤はブレオマイシンが最も効果的なので、ブレオマイシンを(BLM,2.5〜20mg/kg)を単独投与する群と電気パルスと併用する群をもうけた。また電気パルスはElectro Squaree Potator T820(BTX)による矩形波を用い100msecの高電圧パルスを0.5〜1.5KV/cmの強さで1〜10回かけた。腫瘤の大きさを測定するとともに病理学的な観察をおこなって、腫瘍性格が消失するか否かを検討した。更にT820に換えCYU21を用いて低電圧パルス100V/cm以下の電圧で同様な検討を行なった。その結果、通電中は電極の接触した周辺に軽い衝撃が見られたものの、全身性の変化は見られなかった。治療の効果は、肉腫腫瘤9個中9個に細胞浸潤を伴わない壊死を認めた。一方BLM単独投与群では、6個中いずれも全く壊死部分はみられず、肉腫腫瘤の大きさはすべて増加した。またCYU21を用いた低電圧パルス100V/cm以下の電圧でも同様な効果が得られた。このように矩形波を用いたin vivo電気穿孔法により、ブレオマイシンを導入することによってヌードマウスに移植した子官平滑筋肉腫を顕著に縮小することができたことより、電気穿孔法による子宮平滑筋肉腫の治療の可能性が示唆されたと共に、遺伝子導入の条件を見つけ出す素地ができたものと考えられる。
