無歯顎骨窩洞への歯周組織形成誘導に果たすエナメル質基質の役割に関する基礎的研究

研究概要

現在一般に行われている人工歯根インプラントは、インプラント体と顎骨の骨性結合による支持を期待しているが、本研究は人工歯根の支えとして本来の生物学的支持組織(歯周組織)の形成を誘導することを目的とし、本研究期間中にその実現のための生物学的知見を収集することを目指している。近年、エナメル質に歯周組織形成誘導作用があるとの報告がなされ、注目されているが、研究初年度の本年は、エナメル基質蛋白の未梢結合組織に対する生物活性を検証することとし、以下の要領でラット切歯の異所性埋植実験を行った。先ず麻酔下でラット切歯を摘出しエナメル質表面を露出後、同腹ラットの背部皮下に埋入した。経時的に埋入切歯を周囲結合組織とともに摘出し、歯周組織構成要素の形成誘導の有無を免疫組織化学的、酵素組織化学的に検証した。一部の試料で埋植後4週までに幼弱エナメル質表面に骨様硬組織が形成され、切歯舌側の歯根類似部には、神経要素の侵入を伴う歯根膜と歯槽骨類似の構造が確認された。ラット切歯皮下埋入実験系の有用性が確認されたことから、次年度は本実験系における種々の骨関連蛋白および歯周組織特異抗体等の発現経過を明らかにする予定である。
