研究概要	本研究では培養系において単離小脳プルキンエ細胞に対するシナプス形成およびそれに引き続く淘汰の過程のリアルタイム分析を目指している。そこで第一段階として、マウス・プルキンエ細胞の単離培養系の開発を行い、さらに培養系におけるプルキンエ細胞の発達過程を形態学的および電気生理学的に分析した。既存の血清含有培地を用いた中枢ニューロン培養法では小脳プルキンエ細胞を長期間にわたって維持できない。またグリアなど対象外の細胞が増殖しやすい。そこで細胞生存促進因子のうちプルキンエ細胞の生存に必要なものだけを添加した無血清培地(Furuya et al.,1998)を使用し、播種密度などの諸培養条件をマウス・プルキンエ細胞に対して最適化した。その結果、プルキンエ細胞と顆粒細胞を主構成要素とする単純なマウス小脳ニューロン培養系を1ヶ月以上維持することが可能となった。また、この方法により出生直前のマウス胎児から採取したプルキンエ細胞を培養した場合、樹状突起の形態や細胞膜の電気生理学的性質が2〜3間の培養期間を経て成熟することが分かった。これらの成熟過程は生体内におけるプルキンエ細胞の生後発達過程とよく似ている。したがって開発した培養系は所期目的であるシナプス形成を含めて生体内でのプルキンエの発達現象を検討するのに適しているものと考えられる。この成果は、現在論文投稿中である。さらに、生きている培養プルキンエ細胞の形態を観察する技術の確立を目指して、脂溶性蛍光色素(DiIなど)を細胞に直接吸収させる方法やGFP遺伝子導入法を比較・検討中である。
