シナプス前終末の可塑的な細胞内Ca^<2+>遊離機構と伝達物質放出

研究概要	本研究では、シナプス前終末のインパルスによる細胞内Ca^<2+>遊離の活性化機構とその伝達物質放出の可塑性発現機構を調べるのが目的であった。カエル神経筋シナプスとウシガエル交感神経節ニコチン性シナプスに、細胞内微小電極法やCa^<2+>イメージング法やカゴメ化合物活性化法を応用し、以下のことが解った。運動神経終末にライアノジン受容体が存在し、運動神経反復刺激(10〜20Hz、数分)による神経終末内へのCa^<2+>流入により、数分の時間経過で活性化準備(プライミング)され、直ちにCa^<2+>流入により活性化され、インパルスによる細胞内Ca^<2+>上昇を数倍に、伝達物質の放出(終板電位の振幅)を数10倍に促進し、更なるCa^<2+>流入により不活性化され、インパルス停止により数10分の経過で脱プライミングされること、この促進はライアノジンやサプシガーギンにより抑制されること、短期可塑性の内、Facilitationは不変だが、AugmentationとPotentiationが顕著に増大し、その後の促進の減衰は脱プライミングによることが明らかになった。さらに、Ca^<2+>チャンネルとライアノジン受容体と開口放出機構が80nm以内の距離に機能的に連関しており、Ca^<2+>貯蔵オーガネラがシナプス小胞である可能性が示唆された。ウシガエル交換神経節ニコチン性シナプスで、同様の条件刺激により、興奮性シナプス後電位とインパルスによるシナプス前終末内のCa^<2+>上昇が顕著に数時間以上促進され、サプシガーギンで抑制されるがライアノジンでは阻害されないこと、またFacilitationは不変であることが明らかになった。
