研究概要

化学反応系からの複製系の構築、細胞集団の分化、アクチビンによりコントロールされた発生過程の解析、粘菌と大腸菌の共生などの実験を進めるとともに、構成的手法の基盤としてのマイクロファブリケーション設備の立ち上げをおこなった。一方、発生、進化をあるクラスの力学系の普遍的性質として捉える立場からの理論を進めた。各班での進展状況は以下の通りである。(相互に関係しているが便宜上分けて書く)。
*人工複製系構築班
構成的手法により、細胞システム、反応複製系の実験を行ない、特に化学反応系からの増殖、複製、分化機構の創出の実験を進めている。
まず、四方らはDNAを用いた試験管内自己複製系を用い、その再帰的な増殖の出現、そして進化過程を調べた。特に、遺伝子が多くあると、かえって進化性が失われることを明らかにし、そこから情報をになう分子の起源についての実験的解明を行った。これにともない、四方と金子は複製系での情報を担う分子の出現に対する理論を構築している。
ついで、こうした構成的手法の研究をより一般的に進めていくには細胞集団やリポソームの集団をこちらの設定した条件に追いこむための基板が自由にできるのが理想であり、そのための微細加工施設の建設を安田は進めた。既に、神経細胞などをおくための基板の試作品は完成した。その一方でリポソームを合成しつつ電場により集積する基板を完成させた。菅原らの構成した反応合成系と組み合わせて、リポソームの複製実験を始めている。これにはインジェクション装置も組み込んであり、そこに四方らによる反応系を注入する準備を整えた。
一方で、池上らは理論モデルにより、膜を持った細胞要素の形成過程を追っている。また、上の実験に将来対応するであろう反応増殖系の理論解析は金子らにより行われており、同一の細胞がカオス的な反応を持つと分化し、その細胞集団が空間パタンをつくりながら増殖していくことが見出された。今後上のリポソーム実験の進展によって人工的な細胞集団系の構築を目指す際のてがかりを与えている。
*発生過程制御班
浅島らはアニマルキャップといわれる未分化の細胞が、アクチビン分子の濃度の違いにより心筋、脊索、骨格筋などに分化することに既に成功していたが、アクチビン処理したアニマルキャップを時間差をおいて、RA(レチノイン酸)処理することによって、新しい臓器(器官)をつくることに成功した。これは、2つの物質の同時処理と時間差処理では細胞内の記憶に変化が起こることを示しており、理論的に考えられていた細胞内のダイナミクスによる記憶、履歴現象と将来関連しうる結果である。さらに、こうした発生過程での分化と理論での分化ダイナミクスとの対応を考えるには、細胞の性質をセル・ソーター(フローサイトメトリー)により定量的に分離し、細胞内部のダイナミクスや細胞の性質の分布がどう変化して行くかを定量的に追えばよい。そこで、セルソーターを導入し、川戸らと協力して蛍光をつけたデキストランと未処理のもののアニマルキャップの細胞を混合してセル・ソーターにかけて、細胞の分離条件の検討を行った。
一方、四方らは粘菌と大腸菌がある場合に共生関係をつくることを見出していたが、この共生関係が世代を超えて伝わることを明らかにし、その系の時間変化の検討を行っている。これにより、粘菌と大腸菌の共生体がどのような発生過程を持ちうるかの研究を始めている。
理論グループでは、細胞集団の発生過程の理論、シミュレーションを進めた。あるクラスの力学系の振る舞いとして、幹細胞からの確率的分化の機構を与え、細胞が全能性を失い順次決定されていく段階を理論的に明らかにした。また、そのような分化ダイナミクスが生じると細胞が集団として増殖できることを示し、その理由を求めている。このような「理想細胞系」での不可逆な分化は力学系として普遍的に理解されるべきものであるが、その理論は現在模索中である。その点に関連して、佐々らはハミルトン力学系の操作に対する不可逆性の一般理論を提出している。
また、金子と四方は、相互作用による表現型の分化が遺伝型の進化を促すという種分化の理論を提示した。これは実験室で検証可能であり、大腸菌を用いた検証を検討している。
*動的生命解析班
細胞の持つダイナミックなメモリー構造の探索を目指し、準備を進めている。安田らは、心筋培養細胞系の準備を整え、細胞質ヒステリシスの検討を始めている。
一方、測定装置としては局所フラッシュフォトリシス実験装置を製作中、また画像解析データをもとにフィードバック制御を行うための基幹装置としての画像処理システムを立ち上げた。
川戸らは3次元リアルタイム共焦点顕微鏡の開発を行ない、これを用いて浅島らによるアニマルキャップ細胞のCa信号を測定し、今後の発生過程のダイナミクスの研究の準備を行なった。
