研究概要	本プロジェクトは、生物多様性の分子機構を解明し、新しい分子進化学の構築を目的とする。今年度の主な成果◆昨年度の解析をさらに発展させて、3種類の立襟鞭毛虫からチロシンキナーゼ(PTK)mRNAを網羅的に単離し、分子系統解析を行った。その結果、非レセプター型PTKのオーソローグが多細胞動物と立襟鞭毛虫で多数見つかった。一方、レセプター型PTKも立襟鞭毛虫で多く見つかったが、それらはいずれも多細胞動物のものとオーソローグではなかった。この結果は多細胞動物特有の遺伝子の多様化機構の解明に重要な手がかりを与えるものと思われる。さらに立襟鞭毛虫のESTを作成した◆RNA干渉法とDNAチップを用いたプラナリアの脳の解析から、1.脳形成についてはFGFシグナルとその下流に動くMAPK遺伝子群が関与している、2.脳のパターン形成についてはWnt/BMP遺伝子群が関与している、3.脳回路形成にはnetrin遺伝子群が関与していることを明らかにした。◆遺伝子発現に関する多面的なデータの収集を進めた.(DNAマイクロアレイ、in situデータ、他のプロジェクトによる多様な生物種のデータなど)。脳の進化に関しては、プラナリアの遺伝子発現データと脳構造情報を統合して立体的に扱えるようなシステムを他の種のデータも扱えるように開発を進めた。◆1,祖先配列を最尤推定するための効率のよいアルゴリズムの開発、2,複数の遺伝子データを用いて分子系統樹を総合的に評価するためのモデルの開発を行なった。3,これらの方法を用いて、適応的分子進化の検出、真獣類の系統進化、葉緑体ゲノムデータの解析、真核生物の初期進化、ウイルス特にHlVの集団遺伝学的研究などを行なった。◆メダカのトランスポゾンTol-2は内部に自身の転位に作用する転位酵素をコードしている。転位酵素mRNAはexon1〜4から構成される長いmRNAとexon2〜4から構成される短いmRNAの2種類が発現している。そのうち長いmRNAは転位促進にはたらき、短いmRNAは抑制的にはたらくことが判明した。◆顎ロ類とヤツメウナギの口器発生において、相同な分子カスケードが、異なった細胞集団に用いられていることを明らかにした。◆本研究では体表模様の形成に関わる遺伝子hagoromoのmRNAの発現が湖のシクリッドにおいて非常に多様化していることを明らかにしてきた。そこでhagoromo mRNAの機能を調べるためにシクリッド生体の皮下でのhagoromo遺伝子の強制発現による色素細胞の分化を試みた。その結果皮下での遺伝子の強制発現に成功し、現在色素細胞の誘導、分化を検出することを行っている。◆スッポンとシマヘビの染色体地図を作製した結果,ニワトリとスッポン間で非常に高い染色体相同性が存在することが明らかとなった.一方,ニワトリ-シマヘビ間ではニワトリ-スッポン間より染色体相同性は低く,カメ類はヘビ・トカゲ類よりも鳥類とワニ類に近縁であるという最近の爬虫類の分子系統関係を支持する結果となった.
