研究概要	本プロジェクトは、生物多様性の分子機構を解明し、新しい分子進化学の構築を目的とする。本年度の主な成果:◆組織特異的遺伝子の多様化の時期を推定する上で、脊椎動物の高次分類群の系統関係は重要だが、従来のミトコンドリアDNAに基づく分子系統樹は信頼性に欠ける。核DNAにコードされた多数の遺伝子による見直しの結果、これまでの定説と異なり、シーラカンスと肺魚は肉鰭類として姉妹群を形成し、四足動物、条鰭類、肉鰭類の三者がほぼ同時に分岐したとする系統関係を得た。◆MCMC法によるベイズ解析で候補となる系統樹の数を絞り込んだ上で、これらについて最尤法により解析するML法を開発した。さらに、ベイズ法により、分子進化速度が系統樹上で進化するというモデルのもとで、哺乳類や陸上植物の進化における分岐年代を推定し、生物学的に興味深いことをいくつか明らかにした。◆比較染色体マッピングから,ニワトリZ染色体とスッポン6番染色体が高い相同性を示し、鳥類の性染色体がカメ類で高度に保存されていることが明らかとなった。一方、ニワトリZ染色体はシマヘビの2番染色体に対応し、ヘビ類と鳥類の性染色体は起源が異なることが判明した.◆本年度は、主にプラナリアの脳をつくる遺伝子プログラムの解読を行い、脳をつくる遺伝子プログラムが4つのステップから成ることを明らかにし、各ステップに関与する遺伝子プログラムの解読を進めた。◆昨年度までに、顎口類とヤツメウナギの口器発生において、相同な分子カスケードが、異なった細胞集団に用いられ、ヘテロトピーが顎の進化の背景になったことを明らかにした。今年度は、顎の進化において残された問題のいくつかを扱い、顎口類における梁軟骨の起源、三叉神経をはじめとする鰓弓神経系の構築、Hoxコードの進化に焦点を当てて解析した。◆ノックイン技術により作製した3色性マウスにおいて、内因性と変異型の視物質をそれぞれ含む2種類の視細胞からの応答が網膜神経節細胞で統合されていることがわかった。また、脊椎動物の眼の進化過程で、ロドプシンが可視光を受容するのに必須なアミノ酸を新たに獲得したことを発見した。◆前年度までに発見したメダカのTo1-2は脊椎動物で発見された初めての動く能力をものDNA型のトランスポゾンであるが、To1-2因子の構造と機能を解析することで脊椎動物ゲノムの多様化の機構にはたすトランスポゾンの役割を明らかにした。◆脳の進化に関して、プラナリアの遺伝子発現データと脳構造情報を統合して、各種生物間における神経系関連遺伝子の多様化に関する比較進化学的解析を行った。
