磁気刺激および電流分布イメージングによる脳機能ダイナミックスの研究

研究概要

本研究は、脳神経の局所的磁気刺激による脳神経活動の制御、及び新しい手法による神経電流活動の電流分布イメージングや脳磁図・脳波計測により、高時間分解能、高空間分解を有する新しい脳機能ダイナミックスイメージング法を構築することである。
本年度は、磁気刺激時の脳波計測を行い、磁気刺激による脳内信号伝播を調べること。脳磁図計測により、認知活動、記憶活動の脳活動を調べること。及び、MRIを用いた電気情報イメージングの技術を確立し、脳機能ダイナミックスイメージングの要素技術の確立を行った、また、同時に強磁場、およびパルス磁気刺激の安全性、および生体への影響に関しても検証を行った。
脳神経磁気刺激においては、磁気刺激時の磁気ノイズを除去できる脳波計測システムを用いて、磁気刺激によって誘発される脳波の計測を行った。その結果、後頭部と前頭部における興奮の伝播が明らかとなった。脳磁図計測においては、認知反応における注意視野の違いによる脳内処理が、刺激後約200msに後側頭部で生じていることがわかった。MRIを用いて、体内のインピーダンス分布を画像化する方法として、新しく拡散強調MRIを応用した方法を提案し、ラット脳のインピーダンス分布を画像化することに成功した。
パルス磁気刺激の影響が、脳神経細胞にどのような影響があるか、また、磁気刺激の治療効果の可能性を、ラット海馬を用いて調べた。ラット海馬における長期増強現象(LTP)に着目し,TMSがラット海馬に与える影響を検討した。ラット海馬のフィールドEPSPを調べた結果、fEPSPの増強効果や逆に抑制効果が見られ、刺激条件によって異なる結果となった。
さらに、定常磁場の生体影響に関し、再生医学の基礎技術のひとつとして細胞の磁場配向制御法を開発し、神経細胞(シュワン細胞)で磁場による細胞配向制御を実現するなど、強磁場の積極的な医学応用においても成果を得ることができた。
