研究概要

1)可視化プローブの開発
一酸化窒素(NO)可視化のためのプローブについては、NOとの結合が十分強い蛋白質ドメインのより詳細な情報を得る実験に成功し、蛍光物質とのコンジュゲートが完成した。また、リン酸化プローブ作製に向けた基礎的実験成果を得た。具体的には、セリンスレオニンキナーゼによるリン酸化を認識するドメインであるWWドメインを単離し、リン酸化ペプチドとの結合様式を評価した。さらに、NFATの核内移行をモニターするプローブが完成し、その活性化因子である細胞内カルシウムとの詳細な時間的関係を明らかにしつつある。可視化プローブの生細胞への導入法としてシンドビスウイルスを用いる方法を確立した。
2)血管収縮制御機構解明へのアプローチ
血管系をできるだけインタクトな状態で摘出して、細胞内蛍光イメージングを行える実験システムの確立に着手した。
3)神経系におけるシグナル分子の可視化解析
マウス胎児小脳由来のプルキンエ細胞の培養系を確立した。現在、この培養ニューロンに対して可視化プローブを導入し、測定を試みている。さらに、培養系で得られた結果をもとに、生体内の構造が保たれた状態で、生理的条件下におけるシグナル分子の動態を明らかにするため、スライスパッチクランプ用の実験系を立ち上げ、マウス小脳スライス標本を用いた解析にも着手している。
4)カルシウムシグナル系の解析ツール
カルシウムシグナル系で重要な役割を果たすIP3受容体のカルシウム感受性に関わるアミノ酸を分子上に同定した。この結果は、今後カルシウムシグナルフローを解析するうえで重要なツールになる。
