神経細胞特異的ポリオウイルス感染機構と病原性

研究概要

イメージング技術により、神経筋接合部に存在するポリオウイルス(PV)がブンガロトキシンと共局在することを見出した。さらに電顕により、シナプスのPVが小胞に包まれて存在することを示した。軸索輸送に関しては、PC12細胞の軸索内において、ヒトポリオウイルス受容体(hPVR)とPVを持つエンドソームは逆行性に輸送されるが、hPVRの細胞質領域に存在するTCTEL1(細胞質ダイニン軽鎖1)認識モチーフに変異を導入した変異hPVRを使用すると逆行性輸送が観察されなくなった。すなわち、hPVRの細胞質領域とTCTEL1の効率良い相互作用がPVの逆行性軸索輸送に必要であることを示した。
PV感染細胞内では、キャップ構造依存性の翻訳開始が阻害されている(shut off現象)。このshut off現象は、PVの2Aプロテアーゼによる翻訳開始因子eIF4Gの切断によると信じられてきた。このメカニズムに加え、2Aプロテアーゼによる新たなキャップ構造依存的翻訳開始の阻害メカニズムを明らかにした。すなわち、2AプロテアーゼがeIF4Eに結合するとeIF4Eのキャップ結合能が失われることを見出した。
PV感染細胞のプロテオミクスによる解析を行い、少くとも14箇のスポットの量が経時的に増加することを見出した。これらの中には複数の熱ショック蛋白質の他、RNA結合蛋白質hnRNP Kなどが存在していた。これまで知られていないhnRNP Kも含まれている。
