mi転写因子(MITF)によるマスト細胞の分化制御

研究概要	mi転写因子(MITF)はマストの分化を制御する重要な転写因子である。本研究においては、1)種々のMITFミュータントマウスの特徴づけ。2)マスト細胞においてMITFが転写因子に関与する遺伝子の同定。3)MITFの作用機構。4)プロテアーゼ遺伝子の転写という4個の研究項目を設定して研究を開始した。MITFをコードするmi遺伝子座には、人工的につくられたものを含めて多くのミュータントが知られているので、MITF遺伝子の異常とマスト細胞の表現形の関係についてしらべた。mi遺伝子座で最初にみつかったmi突然変異遺伝子をホモに持つmi/miマウスの皮膚のマスト細胞とmi/miマウスの脾臓細胞を培養して得た培養マスト細胞の表現形は多くの異常を示す。最初はmi突然変異遺伝子がコードするMITF(mi-MITF)の転写活性性を失われるためであると単純に考えていたが、mi遺伝子自身のプロモーター領域にトランスジンが挿入されたためにMITFが全く産生されなくなったtg/tgマウスの皮膚マスト細胞あるいは培養マスト細胞の表現形と比べてみると、mi/miマウスのマストの異常の方が程度が大きいことが分かった。mi-MITFは転写活性を欠くのみならず、他の転写因子の転写活性を制御していると考えられる。mi,tg以外の突然変異について、そのミュータントのマスト細胞の表現形がmi/miとtg/tgのどちらに似ているか調べてみると、mi-ce/mi-ceマウスと、mi-ew/mi-ewマウスのマスト細胞の表現形は、mi/miよりtg/tgに似ていた。mi-ce突然変異遺伝子で欠損しているzipper領域の1部、mi-ew突然変異遺伝子で欠如しているbasic領域の1部は、正常MITFの転写活性化能にも、またmi-MITFでみられる転写抑制能にも必須を考えられた。
