グロビン遺伝子群の多型を用いた日本人の起源に関する分子人類遺学的研究

研究概要	日本人の起源を検討するにあたり、北方起源の可能性を見るために韓国、台湾の、また南方起源の可能性を見るためにタイ、マレーシア、インドネシアのサラセミアの変異と、変異に連鎖したハプロタイプおよびフレームワークを検討した。昨年度、既に日本人、韓国・中国人、東南アジア人集団いずれにも見られる変異と、日本人と韓国・中国人集団にのみ見られる変異について解析を終わり、日本人の北方起源のデータを報告した。本年度は南方起源の可能性を検討するために、日本人と東南アジア人集団の変異の検索を進めるとともに、両集団のみに見られ、南方から日本へ移入された可能性があるcodons8/9(+G),codon15(TGG→TAG),IVS-1nt1(G→T),codon35(TAC→TAA)の変異の解析を行った。その結果、codon15(TGG→TAG)と連鎖しているハプロタイプは、日本人では(+----++/FW1)でタイ人では(+-----+/FW3)、インドネシア人では(+-----+/FW3)、(++-++-+/FW3)であることが分かった。またIVS-1nt1(G→T)は日本人では(+-----+/FW3)、タイ人では(-+-++++/FW1)、インドネシア人では(+----++/FW1)と連鎖していた。従って、これらの変異はそれぞれ日本と東南アジアで独立に生じ、起源が異なるものと考えられた。なおコドン35(TAC→TAA)およびcodon8/9(+G)と連鎖したハプロタイプは、それぞれタイ人で(+-----+/FW3)、マレーシア人で(+----++/FW1)であった。しかし日本人では家族のサンプルが得られずハプロタイプを決定できなかった。以上から、日本人の起源をサラセミア変異および多型から検討した結果、東南アジア起源を裏付ける結果は得られなかった。
