マウス半数体精子細胞特異的に発現する遺伝子群の解析

研究概要

精細胞特異的cDNAクローニング(精細胞特異的抗体によるクローニング及び精巣全体から支持細胞を差し引くサブトラクションクローニング)によって精細胞にのみ発現し、他の組織では全く発現の見られない新規の遺伝子が多数存在する事も明らかになってきた。これらの我々が単離した個々の遺伝子についてはそれぞれの機能の解析を進めた。pAP-3ベクターを用いて行う我々のサブトラクションクローニング法では半数体精子細胞に特異的な発現の見られる遺伝子群のクローニングを効率良く行う事が出来、すでに多数のcDNAクローンを単離し、これらの遺伝子産物の精子細胞分化に果す役割を順次解析を行った。さらに残りの遺伝子についても解析中である。
我々は、全cDNAを含むクローンを単離し、そのコードする産物の機能を解析する事をめざしているため、すべて完全長cDNAを単離したうえで解析を進めている。さらにこのライブラリーには既知の精細胞特異的遺伝子も多数含まれており、半数体精子細胞特異的遺伝子をほぼ網羅しているものと考えている。またcomputerに登録されたtestis EST libraryから各臓器のEST libraryをsubtractしgerm cell specific ESTのみを拾い上げ、我々のサブトラクションクローニングで得られた結果と比較してみると、testis ESTにのみ存在する半数体精子細胞特異的遺伝子はほとんど見つからず、我々のsubtraction cloningが如何に優れているかを示している。
これらの遺伝子の解析から、半数体特異的遺伝子のゲノム構造がイントロンを含まないもしくは、単純な構造であること、今まで、半数体特異的遺伝子は少ないと考えられてきたが、かなり多く存在すること、ヒトにおいてもこれらマウスの相同遺伝子が機能していること、などが明らかとされた。今後も、さらに継続しこれら遺伝子の解析を網羅的に行っていきたい。
