光合成同化産物の輸送とシグナル伝達の分子機構の解明

研究概要

スクロース・グルコースを中心とする光合成同化物質の代謝・輸送、またその制御に関わるシグナルの認識・伝連機構の解明を目標とする。最終年度は、これに加えて窒素化合物の輸送機構の研究も行った。
1.イネ葉鞘における横走維管束の形成と通気組織形成(細胞死)の空間的相互関係についての観察を行い、葉の発達段階における形成層の動向に関する実験的知見の蓄積をみた。
In situ hybridization法によるOsSUT1mRNAの分布を調べ、この遺伝子の発現が維管束篩部伴細胞に局在すること、また糖によって制御されていることを明らかにした。
2.イネ単糖トランスポーターOsMST1-5のうち、とくにOsMST3,5を中心に研究を行った。酵母発現系を用いて糖輸送速度・基質特異性の詳細を解析した。OsMST3は、グルコース以外にも広く単糖の輸送が可能であり、またエネルギー依存型のH+共輸送体であることを明らかにした。OsMST3は、生育に伴い細胞壁が肥厚する細胞で特異的に発現し、センス形質転換イネの解析から、細胞壁成分の生合成に関与する可能性が示された。
3.等電点電気泳動と新規活性染色法を組み合わせて、イネ胚における6種類のヘキソースキナーゼアイソフォーム(GK1-3,HK1,2,FK1)の同定を行った。酵素化学的解析よりHK1,2が糖センサー候補であるとの知見を得た。
4種類のcDNAのうち、OsHXK1のみが糖によるα-アミラーゼ遺伝子(RAmy3D)の転写制御に関与することを証明した。
4.α-アミラーゼ遺伝子(RAmy3D)とRab16A遺伝子を用いて、糖と植物ホルモンによる制御の関係ついての研究を行った。
糖とジベレリンのシグナル伝達相互作用を検討する目的でGAMYBの発現制御に関する研究を行った。
恒常的ジベレリン応答性イネ変異体slender riceに関する解析とその原因遺伝子SLR1がGAI orthologueであることを明らかにし、植物の矮化機構を解明した。
